Classification of Renin Angiotensin System Drugs | Raas Drugs PPT | Raas Blockers

Raas Drugs:

Renin angiotensin aldosterone system blockers are anti-hypertensive drugs used in mild to moderate hypertensive crisis.
they exhibit pharmacodynamics action by altering raas system ppt.
on basis of their mode of action these are classfied as under.

Raas Blockers classification on basis of pharmacodynamics:

sub-classes of Raas blockers are:-
  • Drugs inhibiting renin (aspartyl protease) release
  • Renin action blockers
  • ACE inhibitors
  • angiotensin I receptors blockers (at1 blockers).

A)  Renin release blockers:

Rennin found in our body as secreted by juxtamedullary apparatus is responsible for conversion of angiotensinogen to angiotensin I (A potent vasoconstrictive agent).raas system is physiologically active but needs to be inhibited one way or another if pathologically stimulated.
As the name indicates These drugs are responsible for inhibiting the release of   aspartyl protease  from juxtramedullay apparatus in nephron.their inhibitory effect is indirect, means they alter certain other mechanisms, that as a compensatory response aspartyl protease release diminishes.
following drugs are renin release blockers.

B)  Renin action blockers:

these drugs block the cleaving action of renin on pepsinogen.thus inhibiting its conversion to angiotensin I.
following drugs are aspartyl protease action inhibitors:-
  1. alaskirin
  2. remiskirin

C)  Ace inhibitors:

These drugs are specially drug of choice specially in hypertension associated with diabatic nephropathy.
angiotensin converting enzyme (simply ACE) is an enzyme that converts AT1 to AT2 (angiotensin II) which is a more potent vasoconstrictor that at1.
Ace inhibitors stops this conversion and are thus responsible for diminishing a more potent vasoconstrictive agent from taking the stance.
ace inhibitors are famous by their suffix " pril"  as  pril class.
following is a list of pril drugs.
  • captopril (capoten ).
  • lisinopril (zestril ).
  • enalopril
  • foesinopril
  • quinapril

D)  Angiotensin I receptor blockers:

these drugs interct in a simple agonist antagonist fasion with angiotensin I.
on basis of chemical nature they can be  peptides  or non  peptides  as below.
peptides:
currently this class has only sarsalasin.
non peptides:
also known as  sartan class .following are some of the drugs included.
  • losartan (cozar)
  • eprosartan
  • valsartan
  • telmesartan
following is an illustration of mode of action of angiotensin 1 receptor blockers and their clinical uses.
